TRIUMPH is a program of four phase 3 multicenter randomized double-blind studies comparing weekly use of the compound against placebo, alongside diet and physical activity, in over 5,800 participants. It uses an unusual nested design: two of the studies are weight-management trials with two further protocols running inside them, a third is a weight-management trial in people who already carry a cardiovascular diagnosis, and a fourth is a standalone trial for one of those two further protocols. Working with isolated, still-beating right atrial tissue from mice in an organ bath, researchers found the compound made the tissue beat faster, with the effect reaching statistical significance at low nanomolar concentrations and, at the higher concentration tested, coming close to the effect of a standard adrenaline-like stimulant. In separate electrically paced left atrial tissue from the same animals, the compound did NOT increase the force of contraction, though the adrenaline-like stimulant did. Blocking each of the three target receptors in turn showed that only the glucagon-receptor blocker significantly reduced the faster beating; the GLP-1 and GIP receptor blockers did not. The effect survived a beta-blocker but was blunted by an inhibitor of the PKA enzyme, and was amplified by a drug that stops cAMP being broken down. The authors concluded the compound speeds the beating rate in mouse right atria through the glucagon receptor, signalling via cAMP and PKA. They framed the study around the observation that this compound raised heart rate in human safety studies, and listed a direct action on the heart's pacemaker as one of three candidate explanations they set out to test. Retatrutide is a synthetic peptide: a single chain of 39 amino acids carrying a 20-carbon fatty diacid on the side chain of one of its two adjacent lysines, a modification that lets the molecule ride along on blood proteins and stay in circulation far longer than a bare peptide would. What sets it apart from the rest of its family is that it engages three hormone receptors rather than one or two: the GLP-1 receptor, the GIP receptor and the glucagon receptor, which is why the literature calls it a triple hormone receptor agonist. It was developed by Eli Lilly and carries the development code LY3437943. Two of the vendor listings surveyed for this page sell it under the name GLP-3, a label that appears on neither of the registry records cited here and in none of the papers below, all of which call it retatrutide or LY3437943.
Most of this record is human trial data, which is unusual for this catalog. It runs to a phase 1b trial, four phase 2 trials and substudies, one phase 3 trial and a meta-analysis of randomized controlled trials, alongside rodent work and organ-bath experiments on isolated heart tissue from mice and from people. The discovery paper reports that in cultured cells the compound 'shows balanced GCGR and GLP-1R activity but more GIPR activity'
, and a phase 1b trial reported a terminal half-life of about 6 days.
It is investigational and in late-stage development. It has no approved use in the United States or anywhere else, and per the sponsor's own public statement it is available only to people enrolled in that sponsor's clinical trials. What the trials report is not uniformly favorable, and none of it is left out here: digestive side effects were the most frequently reported adverse events throughout; heart rate rose in step with the amount given and peaked around 24 weeks; a nerve-sensation side effect (dysesthesia) was reported in up to 12.5% of the groups given the compound against 0.9% on placebo in the sponsor's phase 3 announcement; and the compound's own body-composition substudy found lean tissue coming off roughly in proportion to total weight rather than being spared. - Supplied as a lyophilized (freeze-dried) powder. Standard laboratory practice for lyophilized research peptides is to hold the sealed vial frozen for long-term storage, commonly at or below -4 F (-20 C), kept dry and protected from light, and to avoid repeated freeze-thaw cycles, which degrade peptide material. The fatty-acid tail on this molecule makes protection from moisture particularly relevant. Let a cold vial come up to room temperature before opening it, because cold glass pulls water out of the air. That is general practice for this class of material, not a claim attributed to any supplier or brand.
- Once it is mixed
- Once it is mixed, keep the vial refrigerated at 36 to 46 F (2 to 8 C), out of the light, and do not freeze it: freeze-thaw cycling forms ice crystals that damage peptide and produces concentration gradients inside the vial. Mixed with bacteriostatic water, the standard window is up to 28 days, the same limit any preserved vial gets anywhere else. That 28 days is a sterility limit, NOT a measured chemical stability window for this compound, and it is never to be written as 'stable for 28 days'. A vial mixed with plain sterile water has no preservative in it and is single use. Peptide solutions are also damaged by vigorous agitation, which shears chains and causes foaming at the air-liquid interface. Nothing beyond storage and handling is given here: no volumes, no concentrations, no routes, no schedules and no preparation of any kind.

- None located, and the coverage of that statement is worth naming: the research pass searched the peer-reviewed and primary sources it could re-resolve and found no published stability assay for this compound at all. The solubility figures for DMSO, PBS, water and acetonitrile that circulate for it, and the receptor-potency (EC50) values that circulate with them, come from chemical-catalog product records rather than from any published measurement, so both are stated as unknown here rather than reprinted behind a caveat. Precise-sounding vendor potency claims of the form 'above 90% for two weeks, 85 to 92% by day 28' have no published stability study behind them and are not reproduced. The one compound-specific property that is documented is a terminal half-life of about 6 days, reported in a phase 1b human trial, which is a property of the molecule in the body and not of the vial.
